8-hydroxydeoxyguanosine mediates cell death of KG-1 by inducing apoptosis

Recently, we have reported that 8-hydroxydeoxyguanosine shows the cytotoxicity on KG-1. In this study, 8-hydroxydeoxyguanosine mediated cytotoxicity is attributable to the induction of apoptosis. The apoptosis was evidenced by increase of sub G1 hypo-diploid cells, DNA fragmentation, and morphological apoptotic body formation. This apoptotic process was associated with down-regulation of bcl-2 and formation of cleaved bid by caspase 8, loss of mitochondria transmembrane potential, cytochrome c, caspase 9 and caspase 3 activation. These findings suggest that hydroxydeoxyguanosine mediated cytotoxicity of KG-1 by the induction of apoptosis.
