Published in Cancer Detection and Prevention 1993; 17(1). Mutagenic substances in urine of rats bearing transplanted nephromaDept of Hygiene & Epidemiology, Univ Medical School, Debrecen H-4012, HungaryMesoblastic nephroma (Ne/De) was induced chemically in newborn F344 rats. The tumor cells has been maintained by serial transplantation in female F344 rats for several years, In order to study the mechanism of tumor progression and metastasis, urine samples were collected from the animals, during the development of the tumor. Hydrophilic and hydrophobic organic fractions of the samples were concentrated on XAD macroreticular resins. Mutagenicity of the DMSO-eluates were studied separately in the Salmonella/Ames test, Enhancing mutagenicity of the hydrophobic (XAD-4) fraction was detected with the tumor development in TA 98 strain of S. typhimurium. "Frame shift" mutagens observed in the urine can be produced by tumor cells and might be involved in the process of progression and/or metastasis. KEY WORDS: urine mutagenicity, Ames test, transplanted tumor, F344 rats. Paper presented at the International Symposium on Genetic Factors in Predictive and Preventive Oncology; Nice, France; March 14-16, 1993; in the section on Genetics Of Lymphoid Neoplasia. |
